So, archaeologists exhumed the suspected remains of Mona Lisa. After carbon dating to confirm the time period, they will then attempt to reconstruct her face. Thus proving Mona Lisa was a woman.

Update: So, it will take a few more years before we see DNA confirmation.
Reconstructing her face won't be possible due to there being no skull in the tomb (due to degradation via flooding).
But, there is documented proof that the person who posed for Leonardo was a woman.
